2013At the NYSBCSemester-long Course: "Cryoelectron Microscopy of Macromolecular Assemblies"This comprehensive course will cover the theory and practice of solving molecular structures by electron microscopy. We will start with optics, sample preparation and a basic mathematical description of diffraction before moving into a detailed exploration of the three main methods of structure determination: tomography, single particle analysis, and 2D crystallography. We will end with a discussion of map interpretation and molecular fitting. Two weeks are devoted to practical sessions giving students hands-on experience with a variety of software packages for image reconstruction. Speaker: Manfred Auer, Ph.D. - Life Sciences Division, Lawrence Berkeley National Laboratory, Berkeley, CA A Tale of Two Communities: Electron Microscopy 3D Imaging Across the Scales: Life is organized at various scales from macromolecular machines residing in their native cell and tissue context, over organelles and subcellular regions to the interactions of cells and tissues. Imaging biological function across the scales requires the integration of multiple imaging approaches, and requires sophisticated sample preservation (through high pressure freezing), EM-compatible tag-based labeling approaches, fluorescence and EM imaging (2D wide-field TEM and 3D TEM and Focused Ion Beam and Serial Block Face SEM), as well as advances in image 3D visualization, segmentation and quantitative analysis (including CAD model-building and simulations). Using a few selected examples from our ongoing research portfolio, for the world of eukaryotes, I will show examples at the organelle and subcellular volume imaging by electron tomography (plant cell wall and hair cell stereocilia), as well as the cellular and tissue level (mammary gland branching morphogenesis and breast cancer progression model in 3D cell culture ). For the world of prokaryotes, I will show that bacterial biofilms display remarkable 2D and 3D organization at the ultrastructural level that underlie physiological microbial community function. Frozen-hydrated sections) in their laboratories. Our latest tricks making CEMOVIS quite easy will be demonstrated. Essential background theory of CEMOVIS will be given and most of the time will be spent practicing high-pressure freezing, cryo-sectioning, and low-dose TEM imaging of vitreous sections. 1 high-pressure freezing machine, 3 state-of-the-art cryo-ultramicrotomes and 1 cryo-electron microscope will be dedicated to the course. The 3-day course is given twice for two different groups of maximum 4 participants each so that every participant can be actively practicing during the whole course. All the participants of the previous previous courses have been successful in producing, collecting and imaging vitreous sections. The Sixth Brazil School for Single Particle Cryo-EM(14 - 26 August 2014)This single-particle data processing school is an intensive hands-on course/workshop, in which theory and practice are tightly intertwined.The spectrum of topics covered suits attendees who are new to the field as well as those for whom the technique is already of growing importance in their research and wish to acquire an in-depth understanding of the methodology. Topics range from data collection and three-dimensional reconstruction techniques to the final interpretation of the cryo-EM maps by (flexible) fitting of X-ray structures. The last days of the meeting will be devoted to advanced topics such as dealing with heterogeneous data sets ("4D" cryo electron microscopy) and high-resolution refinements. The attendees are expected to bring along a modern notebook computer onto which all necessary software will be installed. Most processing will take place in the context of the IMAGIC 4D software system but other software packages such as Frealign will also be used. As in the past, the course will provide in depth theoretical and practical training, with the focus on image processing for 3D reconstruction. Registration is now open. The hand-on workshop on the processing of images from cryo-electron microscopy of 2D crystals of membrane proteins (electron crystallography) will provide the theoretical background and practical applications in advanced image processing algorithms. A Workshop on Advanced Topics in EM Structure DeterminationThe National Resource for Automated Molecular Microscopy Announces A Workshop on Advanced Topics in EM Structure Determination November 11-16, 2012 National Resource for Automated Molecular Microscopy The Scripps Research Institute (TSRI), La Jolla, California. We invite applications to participate in a Workshop on EM Structure Determination of Challenging Macromolecules. The workshop is aimed at students, postdocs and faculty that already have some experience practicing cryoEM. We can accommodate a total of 100 participants. A major goal of the workshop will be to discuss some of the challenges that we face in using cryoEM to solve the structure of macromolecules. Topics to be discussed will include improvements in specimen preparation, imaging, processing and reconstruction, and methods for validation. The basic format of the course will be theoretical lectures in the mornings followed by practical demonstrations and panel discussions in the afternoon. Evenings will be reserved for poster presentations by participants and research talks by instructors.
