Low Dose in Serial EM

Setting up Low Dose in Serial EM

  • Search mode
    • 10kx center beam FLC off
    • Expand beam
    • Click SA Diff
    • Use PLA to move Beam to camera
    • Drag FLC to top

* Focus Mode

    • 200kx
    • Check additional beam shift box in Serial EM
    • Move beam with image shift
    • Uncheck additional beam shift box when done moving

  • Obtaining Live focus mode
    • Drop screen in focus mode
    • Set mag/brightness
    • Check/uncheck continous update of mag and beam
      • This remembers: Mag/birghtness/spotsize
    • Switch to TEM mode
    • Lift screen
    • Click unblank in Serial EM

Using Serial EM in Low Dose mode

  • Search Mode
    • Click SA Diff on TEM - it does not remember diffraction
    • Slide FLC
    • Switch to CCD
    • FLC - all off
    • Can take a view image if the other modes are set up
    • If you apply an image shift, click reset image shift to move stage to new position

*Focus Mode

    • Select Focus as area to show when screen down
      • lower/raise screen
    • Switch to TEM
    • Click unblank in Serial EM
    • Focus
    • Switch to CCD
    • Click record

  • Drop screen to change modes
  • Set ALLOWTOPICVIEW =
