Calibrations must be done in admin mode. To save calibrations you must check next to admin in pull down menu. DO not calibrate unless authorized.
Binning- adds up selected number of pixels and makes them 1. Reduces info. Shortens exposure.
Focus area- move to bottom of screen so the camera charge will clear.
16 Bits- camera reads 0-65000. Prefer to truncate data, anything over 32500 is thrown away.
Low Dose- when Tecnai is in exposure mode open serial EM low dose and check box. Focus and tracking should be in the same place. The beam in these areas should not overlap record area. Do not normalize beam through view.
Start with a negative tilt and go positive. Nudge goniometer up higher to avoid hysterisis.
