New User Setup Tecnai F20

  • Create new accounts on both computers and Greymatter
  • Set passwords to never expire
    • Windows 2000 or XP
      • Must be Administrator
      1. Right-click My Computer
      2. Click Manage
      3. Click to Expand Local Users and Groups
      4. Click Users
      5. Double click account
      6. Select Password Never Expires checkbox

  • Set up new users as “TEMexperts” to insure functionality
    • advanced users should also be Power Users. Instructions for multiple groups:
      • Manage Right click My computer --> Manage --> local Users and Groups --> Users--> (highlight name) --> Properties --> "Member of" tab -->Add...

  • Remind user to logout and not "shutdown" computers

New user Low Dose setup

    • make sure specimen rod is still inserted
      • This will make spot size 2 the default, if rod is out 5 is default.
    • Load known good Low Dose file listed on whiteboard
    • save as new user file and folder

  • Prompted upon opening:
    • User name (first initial and last name)
    • Working directory – P: /
  • To place scalebar:
    • scaling/scalebar check "use Scalebar" box
  • 8 bit Tiff

Fast Scan Camera Computer

  • Turn off auto screen off - right click screen: properties\screen saver\power
  • Make background black

Fast Scan Camera:

  • Select “firewire” when prompted upon opening camera
  • Set speed to 300ms
  • To enable flat correction - File\preferences\misc - change C:\program files\... to C:\\tcl\\EMMENU\\tem1000
  • Check flat correction box

Serial EM

  • Use C drive
  • Personal directory
  • Settings are local
    • look feel, camera, tilt series
  • Make a shortcut
    • Right click icon to make sure *Start in : Change to user directory * Target is correct
      • Okay to adopt others settings

Calibrations need to change start in line under properties to prevent over writing

  • Load alignments for F20 and assume the calibrations for serial EM are valid.
    • Stress that it is a bad idea to fiddle with alignments
    • FEG registers Alignments

  • Calibrations must be done in admin mode. To save calibrations you must check next to admin in pull down menu. DO not calibrate unless authorized.

  • Binning- adds up selected number of pixels and makes them 1. Reduces info. Shortens exposure.

  • Focus area- move to bottom of screen so the camera charge will clear.

  • 16 Bits- camera reads 0-65000. Prefer to truncate data, anything over 32500 is thrown away.

  • Low Dose- when Tecnai is in exposure mode open serial EM low dose and check box. Focus and tracking should be in the same place. The beam in these areas should not overlap record area. Do not normalize beam through view.
Start with a negative tilt and go positive. Nudge goniometer up higher to avoid hysterisis.
