December 2013 - X261 CysZ PA7 - Initial screen with P07 strain

Conditions:

  • pH 5.5 or 6.5 + 5mM >nop>NaCl
  • no additive, or 5mM Molybdate or 5 mM Sulfate
  • DOPG:brain-OG or Cardiolipin-DM
  • udialysis plate @37C for 1 week

July 2013 - X212 CysZ PA01 - Initial screen from Idiomarina screening

Protein acquisition form: Protein_acquisition_form_CysZpa_070113.doc.
Conditions inspired from tubular crystals of the Idiomarina homolog:
  • pH 5.5, 6.5 and 7.5 + 50 mM NaCl
  • DOPG:brain-OG at all pH, and at 1 pH: Cardiolin, E.coli, DMPC.
  • Dialysis. Also, a few conditions with biobeads to try sucrose gradient+ANS.

X212, Summary

Image selection: X212-CysZPa-ImageSelection.pptx. Crystalline patches in DOPG:brain-OG, pH 5.5, LPR 0.75-0.9 (same lipid, same LPR as Idiomarina). No pattern at higher pH or other lipids tested.
