Tecnai Tomography

Microscope Alignment

  • Critical to proper functioning of tomograpy
  • Set specimen to eucentric height
  • Gun alignments (tilt and shift)
  • Beam alignments (rotation center, pivot points, coma free)
  • Img alignments (pivot points)
  • Skip cosmetic alignments and calibrations

Camera Configuration

  • Incompatible with TCL and EM Menu
  • Start up TIA
  • Supposed to coordinate low dose through Tecnai Low Dose setup
    • so start Low Dose
  • Switch camera control box to CCD

Microscope operation

  • Start FEI tomography package
  • Select TEM Tomography component (In Experiments Box)
  • TEM Basic Calibrations press start
    • auto eucentric height
    • optimized holder position
    • precalibration curve
  • Make sure all 4 calibration button are green.
    • calibrations should be valid from previous run unless change camera or goiniometer
  • Select TEM Auto Functions in Experiment box
    • In Eucentric height Uncheck box calling for lower mag
  • Run TEM Tomography
  • Make sure both LED's are green
    • will have to calibrate focus each time (depends critically on alignments)
  • Go to Acquire tilt series and set up desired parameters
